<font face="Verdana"> </font> <p style="MARGIN-TOP: 0px; MARGIN-BOTTOM: 0px" align="justify">Surface plasmon waves in the visible range are not optimal for studying living cells because the cells' size considerably exceeds the penetration depth of visible-range surface plasmon waves. Infrared (IR) surface plasmons, on the other hand, penetrate much deeper and are more appropriate for studying living cells. The infra-red wavelength is of the order of the cell size. We demonstrate here a mid-IR SPR technique that is based on a gold-coated ZnS prism and uses a Fourier-transform infrared (FTIR) spectrometer as a light source. This is a real time and label-free method to study morphology, thickness, spreading and cell-cell interactions. We also consider surface plasmon propagation in a planar array of living cells, in particular, surface plasmon propagation length. We demonstrate also guided optical waves in a living cell culture as an excellent tool to determine the cell thickness. The surface plasmon measurements are complemented by optical microscopy imaging. We show how highly sensitive surface plasmon technique allows to track the dynamics of different phases in epithelia formation, from individual cell spreading to continuous monolayer. The technique is sensitive enough to measure physiological concentrations of glucose in water and in human plasma. It allows real-time and label-free studies of lipid membranes and cells cultured on the gold-coated surface, particularly drug and protein penetration into cells.
